中文摘要本论文着重研究固定化青霉素酰化酶生产6-APA的技术,为解决我国半合成抗生素的中间体问题而努力,并推动国内固定化酶技术的研究向工业应用过渡。一、理论分析了青霉素酰化酶的反应动力学和分子作用机理,建立起新的动力学方程,比以往的方程更接近实验测定结果。二、研制新型的固定化载体。研制了一种复合载体,以之制备的固定化青霉素酰化酶的活性较好,步骤较为简单,载体成本低。三、测定了新型固定化青霉素酰化酶的性质和动力学常数。四、计算了底物浓度和各种抑制作用对反应过程的影响及连续反应过程,结果表明三、四级连续反应效率较高。本工作始于1994年,1996年申请国家“九五”攻关获得批准(与微生物所联合承担)。

英文摘要This work aims at developing new technologies of immobilized penicillin amidase to solve the problem of emergent demand for 6-APA in producing semi-synthetic antibiotics and enhancing the transition of researches on immobilized enzymes (IMEs) to industrial applications. First, the kinetics and molecular mechanisms of penicillin amidase are discussed theoretically. A novel equation is deduced, the predictions of the novel equation agree better with the experiment data than former equations. A novel reasonable molecular mechanism of penicillin amidase is established and the essential relationship of the novel equation and the Warburton equation is discussed. Second, novel carriers are studied for enzyme immobilization. A novel composite carrier was developed. Third, the properties and kinetic parameters of the novel immobilized penicillin amidase is determined. Finally, calculations were carried out on the effect of the substrate and the products on the enzymatic reaction and on continuous reactions. The results show that three- or four-stage continuous reactions have great potentiality. This work began in 1994 and got the support of Ninth-National-Five-Year-Program at the end of 1996.
