中文摘要酿酒酵酒(Saccharomyces Cerevisiae)是一种重要的发酵工业和生产基因重组药物的菌种。本文从单细胞水平上研究了酿酒酵母的生长与代谢。本文确定了一套高密度同步化培养醇母细胞的方法。用不连续Ficoll400密度梯度离心法筛选得到同步化细胞,然后进行酵母细胞的同步化培养。同步化指数SI为0.81,细胞密度达10~7Cell/mL。用上述同步化培养的方法研究了酵母单细胞的生长代谢行为。发现在酵母细胞生长过程中,葡萄糖的吸收速率和乙醇的产生速率在整个细胞周期中是不同的。葡萄糖的吸收峰和乙醇的产出峰出现在细胞分裂前的1-1.5小时之内,当葡萄糖吸收快的时候。乙醇的产出量也大。目前尚不能确定两个峰出现的先后顺序。根据对照实验,可以肯定,这些实验现象不是由同步化方法本身造成的假象。在实验的基础上建立了一个酵母单细胞生长代谢模型。模型引入了体内ATP水平对细胞生长代谢的调控作用,描述了细胞个体从正常个体的1/8到出芽生殖出一个子细胞过程的生长代谢。模型结果与实验结果以及文献报道的结果吻合得比较好。
英文摘要Saccharomyces Cerevisiae is an important strain for fermentation industry and producing gene-recombined drugs. In this work, the growth and metabolism of a single S. Cerevisiae cell was studied. A practical method for synchronous culture with high cell density was established. Synchronous cells are selected by discontinuous Ficoll400 density gradient centrifugation and then are cultured synchronously with density of 10~7cell/mL. The synchronous index(SI) reaches 0.81. The growth and metabolism of a single S. Cerevisiae was studied by this synchronous culture method. The consumption rate of glucose and the production rate of ethanol vary with the cell cycle and exhibit periodic behavior through the culture. The consumption peak of glucose and the production peak of ethanol appear within 1-1.5 hours before the cell splits. The appearing sequence of the two peaks can not the determined at present. According to the controlled experiments, these phenomena are not the artifacts caused by synchronous culture method. A single cell model of S. Cerevisiae was constructed based on the experimental data. The regulation of ATP on growth and metabolim was introduced to this model. The model describes the growth and metabolism process, that is, from a justly splited bud to the next division. The predicted behavior can fit the experiments.
