Multilayer hydroxyethyl cellulose-adsorbed coating on poly(dimethylsiloxane) microchips for protein separation

英文摘要In this paper, we present a fast method for generation of hydrophilic surface coating for poly(dimethylsiloxane) (PDMS) microchip. The native PDMS surface was modified by multilayer-adsorbed and heat-immobilized hydroxyethylcellulose (HEC) after hydrogen peroxide solution treatment. Repeating the coating procedure three times was found to produce the most effective coating. The three-layer HEC coating converts the original PDMS surface from a hydrophobic one into a hydrophilic surface, and suppressed the electroosmotic flow (EOF) in HEC-coated channels over a wide pH range of 3-11. Four fluorescently labeled enzymes derived from multienzyme tablet were separated satisfactorily on the HEC-adsorbed PDMS microchip. Good separation of standard proteins based on molecular weight was also obtained on the coated chip.
