Potent anti-inflammatory activity of polysaccharides extracted from Blidingia minima and their effect in a mouse model of inflammatory bowel disease

英文摘要Blidingia minima is a green alga that is ubiquitous on many coastlines. To explore the potentially beneficial effects of polysaccharides extracted from Blidingia minima (BMP), in vitro and in vivo studies were performed using dextran sulfate sodium (DSS)-treated intestinal epithelial cells (IPEC-J2 cells) and mice with DSS-induced colitis, respectively. BMP showed an anti-inflammatory effect on DSS-treated cells. Furthermore, BMP alleviated colitis symptoms in DSS-treated mice. BMP repaired colonic dysfunction, and colonic morphology, infiltration, and the expression of tight junction proteins were all improved. Moreover, the mRNA expression and level of pro-inflammatory cytokines, as well as the phosphorylated protein levels of AKT, I kappa B-alpha, and NF-kappa beta in colonic tissue, were all decreased by BMP supplementation. We suggest that the anti-inflammatory effects of BMP may be associated with the modulation of NF-kappa B pathways. These results promote the potential application of BMP as a novel, next-generation therapeutic for treatment of inflammatory bowel disease.
