英文摘要Cell ablation is a classic approach to understand cell and tissue function in development or neuronal function in vivo. mini singlet oxygen generator(mini SOG), engineered from the LOV domain of Arabidopsis phototropin 2, was originally developed for correlative light and electron microscopy(Shu et al 2011 PLOS Biol). After illumination with blue light, mini SOG generates the reactive oxygen species singlet oxygen(1O2). Illumination of neurons expressing mini SOG targeted to the outer mitochondrial membrane(mito-mini SOG) causes neuronal death by a non-apoptotic mechanism, allowing photo-inducible cell ablation(Qi et al 2012 PNAS). Since its development as a cell killing tool, mini SOG has been used in a variety of ablation experiments, using blue light exposures of 0.5-1.5 h. However, extended blue light exposure itself can be deleterious to C. elegans.
