英文摘要干细胞研究因其重要的理论意义和广泛的应用前景而成为近年来生命科学研究的热点领域. 目前人们在胚胎干细胞的定向分化和诱导多能干细胞细胞系的建立方面取得了重大突破,但对相关生物学过程的调控机制所知十分有限,干细胞应用仍然面临许多难题. 表观遗传调控作为整合细胞内外环境因素与基因组遗传信息的媒介,直接参与控制基因表达,决定细胞分化与功能特异化,在干细胞的自我维持和定向分化以及体细胞重编程过程中发挥了非常重要的作用. "干细胞编程与重编程中表观遗传调控的分子机制和结构基础"国家重大科学研究计划项目围绕干细胞编程与重编程过程中的表观遗传调控所涉及的关键科学问题,利用多学科的研究方法和手段开展研究,目前已经取得了一批具有重大国际影响的突破性成果. 例如,在国际上首次通过单细胞基因组学分析方法,实现了对人早期胚胎的DNA甲基化调控网络的系统分析;又如,利用染色质体外组装体系和冷冻电镜三维重构方法,在国际上首次解析了30 nm染色质纤维的高分辨率结构,在染色质高级结构这一重大科学问题上取得了突破性的进展. 这些研究成果有助于深入理解人类胚胎干细胞及iPS细胞自我维持、定向分化、重编程等各个环节的表观遗传调控机制,将有力地推动基于干细胞的各种再生医疗技术和手段的应用进程.; Stem cell research has been a very hot frontier re-search field in life sciences due to the scientific impor-tance and the great potentials in clinical applications . Although breakthroughs have been made in the directed differentiation of embryonic stem cells and in the estab-lishment of induced pluripotent stem cells , the knowl-edge about the regulation mechanisms of the biological processes is very limited , putting hurdles in the way of clinical usage of stem cells .It has been well known that epigenetic regulation integrates the genetic information and the intrinsic and environmental cellular signals and thus plays important roles in eukaryotic gene expression and in programming and re-programming of stem cells . This project focuses on the functions of epigenetic regu-lation and the underlying molecular mechanisms in the programming and re-programming of stem cells .Here we report the recent breakthroughs of the project , inclu-ding revealing the critical features of the methylome of human early embryos using a single-cell reduced repre-sentation bisulphate sequencing technique and determi-nation of the high-order structure of 30 nm chromatin fi-ber.These studies have greatly advanced our knowledge in fundamental biology related to human health and dis-eases and will accelerate the development of regenerative medicine and biotechnology based on stem cell biology .; 干细胞研究国家重大科学研究计划; 中国科技核心期刊(ISTIC); 4; 24-29; 17
