多级结构表面调控干细胞成骨分化机理的研究

英文摘要The loosening of implants from bone tissues has been a cause of problems in reconstructive surgery and joint replacement, which is resulted from failed osseointegration. Osseointegration is the consequence of a cascade of molecular and cellular events that occur after placement of an implant. Considering cellular functions can be controlled by the interactions between cells and biomaterials, the surface chemistry and topography of support materials play a pivotal role in modulating cell behaviors. In this study, a special hierarchical micro/nanotopography is taken advantages to regulate human mesenchymal stem cell differentiation. The aim is to figure out whether the special surface topography can promote osteogenic and chondrogenic differentiation and the related mechanism, and to evaluate whether better differentiation results can be acquired without chemical compound addition. First, MSCs were cultured on different hierarchical micro/nanotopography surfaces in differentiation medium, and Alizarin Red staining and Alcian blue staining was performed to screen surfaces with higher differentiation efficiency. Then, biological technique including qPCR, RNAseq and CRISPR/Cas9 technology will be employed to analyze the mechanism that hierarchical micro/nanotopography regulates MSCs differentiation. The results by now are limited, but the continued work will help the development of tissue engineering by improving osseointegration of surface modulated implants.
