英文摘要 | Cancer has been becoming one of major threats to human health.1Development of highly integrated strategies for precise diagnosis and efficient therapy of cancer has attracted much attention in the past decades. Imaging-guided therapy as a novel theranostic strategy demonstrates an enhancement in location of tumor site, detection of treatment process and evaluation of therapeutic efficiency. The design and preparation of theranostic nanoagents that integrate imaging and therapy functionalities into a single nanoplatform hold a great promise to enhance the accuracy of diagnosis and improve the efficacy of therapy.Herein, we develop a theranostic nanoagent based on indocyanine green(ICG)-loaded polydopamine (PDA)-iron ions coordination nanoparticles(PDA-Fe3+-ICG NPs), which is used for photoacoustic (PA) and magnetic resonance (MR) dual-modal imaging-guided cancer PTT treatments. In this nanoplatform, ICG molecules, the U.S. Food and Drug Administration approved near-infrared (NIR) dye, absorb PDA NPs (a melanin-like biopolymer) to significantly increase the NIR optical absorption of PDA NPs nearly 6 times and decrease their fluorescence emission, which could improve PA contrast ability and promote photothermal conversion efficiency of PDA NPs. Meanwhile,Fe3+ ions chelated on PDA NPs offer T1-weighted MRI contrast agent (r1=14 mM−1s−1). In mouse 4T1 breast tumor model, the PA/MRI dual-modal imaging and highly efficient PTT treatments with low laser density are achieved with remarkable therapeutic efficiency and minimal side effects. This study illustrates that the highly integrated and biocompatible PDA-based NPs could serve as a versatile nanoplatform with loading different imaging molecules and drugs for multi-modal imaging and cancer combination therapy. |
