Puerarin Prevents Ovx-Induced Bone And Skeletal Muscle Loss In Rats

英文摘要Introduction: Puerarin is an isoflavonoid isolated from the Pueraria Labata (Willd.) Ohwi for treating coronary heart disease as an adjuvant drug in clinical. Our previous study found that puerarin promoted osteogenesis and inhibited adipogenesis in vitro. Objectives: In this study, we are going to investigate the effect of puerarin on preventing osteoporosis in vivo. Materials and Methods: Fifty 6-month-old female SD rats were weighted, and then divided into either sham-operated (Sham, n=10) or OVX operated such as OVX(n=10), estradiol treatment group (n=10), low dose puerarin (LP, n=10), and high dose puerarin (HP, n=10) randomly. Both the Sham and OVX rat were treated with 0.5% CMC-Na, the vehicle used to suspend puerarin and estrodiol. Body weights were recorded every week. All rats were injected subcutaneously with 1% calcein green (10mg/kg) in a time sequence of 13, 12 and 3, 2 days before euthanasia for studying bone mineral apposition. After euthanasia using pentobarbital sodium (120 mg/kg body weight) at the end of 12-week treatment, blood sample was collected from heart for serum isolation, proximal tibiae and vertebrae were dissected for measurement of trabecular micro-architecture, the left femur was subject to biomechanics. Results: Body weight of LP and HP were significantly lower than the OVX group (P < 0.01), and had no differences with Sham group after 8 weeks. Both uterus weight and epithial width of two puerarin groups were significant higher than OVX but lower than E2 except that the epithial width of LP had no difference with OVX. Estrodiol increased by 1.96 folds (P < 0.01) and there is slight increase(0.28 0.52) with significant different between any dose of puerarin with OVX group. Puerarin group showed dose-dependent in results of proximal tibia metaphysis trabecula bone (BV/TV, Tb.N, Tb.Sp). HP group had significantly higher than OVX, but LP not. Compared to sham group, parameters of micro CT (BV/TV, BMD, Tb.N, Connective density) in high dose group of puerarin all increased, conversely, parameters such as SMI, Tb. Sp in high dose group all decreased. Estradiol and both does of puerarin were able to keep the soleus and gastrocnemius weight percent (P < 0.05). Conclusions: High dose puerarin can prevent bone loss and amyotrophia in OVX-induced osteoporosis in Rats. It also showed the effect of phytoestrogen which was safer than E2.
