Optofluidic devices for single-step miRNA detection using molecular probes

英文摘要MicroRNAs(miRNAs) have attracted extensive interest as novel biomarkers in diagnosis and targeted treatment of many diseases, and quantification of miRNAs can be potentially useful for early disease detection, surveillance monitoring and drug response evaluation. Because of the short length, high sequence similarity and low abundance of miRNAs in vivo, it is challenging to realize the sensitive and selective detection of miRNAs with conventional methods. Here, we developed an innovative fiber-based optofluidic platform to perform miRNA identification and quantification using molecular probes. The optofluidic device integrated optical fibers and on-chip microlens, and the compact design increased the detection sensitivity and eliminated time consuming optical alignments. First, we demonstrated the on-chip miRNA detection using fluorescence resonance energy transfer (FRET) principle. A three-way junction assembled FRET probes were encapsulated in droplets to carry out ratiometric miRNA detection with highly selective discrimination of single-base mismatch. To improve the sensitivity of the detection system, we introduced another molecular probe in the optofluidic system using bioluminescence resonance energy transfer (BRET) principle, in which the donor fluorophore is replaced with a self-illuminating luciferase. To carry out the miRNA detection steps, the optofluidic device integrated a variety of components, such as droplet pico-injection unit and high performance fiber-based optical detection unit, and single-step miRNA detection can be performed with simple device operation. In summary, the compact optofluidic system provides a low-cost and highly efficient approach for miRNA identification and quantification with high sensitivity and specificity, and has great potential in point-of-care clinical diagnostics.
