中文摘要chitin is a natural polysaccharide, which consists of a copolymer of n-acetyl-d-glucosamine and d-glucosamine residues, linked by -1,4 glycosidic bonds. chitosan is the deacetylated form of chitin,the major component of exoskeletons of insects and crustacea, and can also be found in the cell walls of some fungi. oligochitosan is the fragments of chitosan, prepared by enzymatic hydrolysis,with higher elicitor activity and water solubility than chitosan, applied as an effective environmentally safe means for plant disease control (kim and rajapakse, 2005). in the present work, a hplc procedure utilizing reversed-phase chromatography coupled with a coulometric array detection system was developed for the characterization of severalmajor secondarymetabolites in tobacco (schmelz et al., 2003). in a single step, leaves are extracted in aqueous acidifiedmethanol, and the extractedcompounds are separated by reversed-phase hplc with an acidic water/acetonitrile gradient in 65 min. the utility of the method in quantifying changes in the secondary metabolites of tobacco after oligochitosan treatment, a treatment known to elicit resistance to herbivores in nature, is illustrated (keinanen et al., 2001). oligochitosan treatmentelicitedincreases insomesecondarymetabolites (chlorogenic acid, p-coumaric acid and nicotine increased 1.5-, 4.6-, and 2.4-fold, respectively) but left others, caffeic acid and scopoletin, undetected in both control and treat group, and rutin, changed with no significance. such broad-based analytical screens will help characterize environmental changes in secondary metabolite profiles and illustrate mechanism of oligochitosan-induced resistance.
