中文摘要microorganisms that can accumulate lipids over 20% of their biomass are defined as oleaginous microorganisms [1]. some yeast strains, such as rhodosporidium sp., rhodotorula sp. and cryptococcus sp. can accumulate intracellular lipids as high as 70% of cell dry weight [2]. to produce microbial lipids using lignocellulosic material-based carbohydrates, we have collected over 30 oleaginous fungal strains in our lab. in the present study, the sequences of the 26s rdna d1/d2 domain and the internal transcribed spacer 1 (its1)-5.8s rdna-its2 region were used to differentiate species in the oleaginous fungi. the corresponding rdna sequences were amplified using d1/d2 forward primer / d1/d2 reverse primer and its forward primer / its reverse primer obtained from d317 fungi identification pcr kit, respectively. the pcr products were purified, and sequenced by takara biotechnology (dalian, china). taxonomic study was performed using blast against fungal sequences in known dna databases [3] and multiple sequence alignments by dnastar software, carried on the molecule statistics analysis by clustal w method [4]. the phylogenetic tree was inferred by treeview software. results showed that strains of our oleaginous collection are distributed in species of rhodosporidium sp., rhodotorula sp., cryptococcus sp., lipomyces sp., yarrowia sp., trichosporon sp., endomycopsis sp., and geotrichum sp.
