中文摘要 | an anti-diuron immunoaffinity extraction column was fabricated for the first time by encapsulation of anti-diuron monoclonal antibodies in sol-gel glass matrix. the experimental capacity of the sol-gel immunoaffinity column (sg-iac) immobilized 240 g antibodies was 302.1±11.6 ng (n=4) of diuron, corresponding to 40% of the theoretical maximum capacity. the antibody density in sg-iac up to 3.75 mg g-1 resulted the linear increasing of experimental capacity. the immunoaffinity column-based loading, rinsing and eluting processes were optimized. based on these optimizations, the sg-iac selectively extracted diuron from an artificial mixture of 64 pesticides. in addition to this high selectivity, the sg-iac proved to be reusable. the sg-iac was combined with liquid chromatography-tandem mass spectrometry to determine diuron in surface water, and the linear range was up to 2.2 g l-1 with correlation coefficient higher than 0.99 and relative standard deviation lower than 5.7% (n=8). the limit of quantitation for 25-ml surface water sample was 3.9 ng l-1. |
