中文摘要	dissociation constant is a very important physical chemical parameter to the compounds. it contributes to an understanding of many phenomena such as reaction rates, biological uptake, transport, environmental fate. there are many methods to determine the dissociation constant, titration, potentiometry, liquid-chromatography, spectrophotometry, capillary electrophoresis and so on. in the last twenty years, liquid chromatography was used to determine this parameter due to its advantages such as small quantity of the analyte required, no requirement of the high purity of the sample. in this paper, a new method was developed to determine the pka value of compounds using gradient elution system with uplc. the experiment proceeded by means of a rp18 which can reduce the interaction between the analyst and solid phase. a binary mobile phase system under gradient elution conditions was used which consisted of triethylamine- phosphate buffer solutions of different ph values and acetonitrile with 40% triethylamine- phosphate buffer solutions. five times linear gradient experiment was used by software csass to calculate the a, c values of the target compound. after having a, c values, the predicted retention time (rt) can be obtained in any chromatographic conditions. pka was the inflection point of sigmoidal curve which reflected the dependence of the analyte capacity factors upon the ph of the mobile phase. the pka of tetrahydropalmatine have been determined, and it also predicted the pka of berberine, palmatine which were known a strong bases. the result demonstrated that this method was simply and time-saving.
