Impact of the intestinal metabolites of ginsenosides on human cytochrome P450 enzymes

中文摘要the intestinal metabolites of ginsenosides rather than naturally occurring ginsenosides are really responsible for the main pharmacological activities of ginseng, but the knowledge concerning the effect of those on human metabolism enzymes is still lacking. we found that the intestinal metabolites of ginsenosides demonstrate a wide range of inhibition of cyp-mediated metabolism and in some cases exhibit activation rather than inhibition. it is noteworthy that compound k (c-k), protopanaxadiol (ppd), and protopanaxatriol (ppt) all exhibit intermediately inhibitory activity against cyp2c9 activity, and ppd and ppt also exhibit potent inhibition of an enzyme-kinetically competitive type against cyp3a4 activity. further study showed that the inhibition on cyps is not due to mechanism-based. the results suggest that the ginseng-derived products might influence cyps activity via their intestinal metabolites, which might be a noteworthy new pathway resulting in clinically significant drug-drug interactions.
