Visual Tracking of Enterovirus Infection Behavior by Bioorthogonal Near-infrared Quantum Dots

英文摘要Enterovirus 71 (EV71) is the main causes of hand-foot-and-mouth disease (HFMD). Recently, EV71 infections were reported to cause high fatalities and severe neurological disease and pulmonary edema in children. However, the mechanism of EV71 pathopoiesis is still unclear, and no effective treatments have been developed. In this study, we report a bioorthogonal strategy to high-efficiently label EV71 for in vivo tracking virus infection with near infrared quantum dots (NIR QDs). The conjugation of QDs onto EV71 was highly efficient with good stability both in vitro and in vivo. QDs labeled EV71 (QD-EV71) maintained viral infectivity and replication capability and their infection behavior is consistent with wide-type virus. Virus infection was monitored in host cell and in living mouse by measuring QD fluorescent signal, and we can visualize viral infection in a noninvasive and real-time manner. Furthermore, QD-EV71 demonstrated bright and sustained fluorescent signals in brain, the lung and gut. The results suggested that EV71 induced pathological behaviours were governed by viral tissue tropism and distribution, and virus mainly concentrated in central nervous system, the lung and gut tissues. Hence, virus labeling with NIR QDs provides a simple and reliable strategy for real-time visual tracking EV-71 infection behaviour in vivo.
