英文摘要 | Objective: Recombinant human platelet-derived growth factor (rhPDGF-BB) is a clinically proven growth factor with the potential of promoting cell proliferation and osteogenic differentiation during the bone regeneration process. This study was to prove our hypothesis that rhPDGF-BB was able to prevent bone mass loss in a steroid associated osteoporosis (SAOP) rabbit model, meanwhile we employed the mouse embryonic mesenchymal stem cells C3H10T1/2 to investigate the mechanism of rhPDGF-BB for treatment of SAOP. Methods: SAOP was induced in rabbits by injection of liposaccharide (LPS) and methylprednisolone (MPS) according to our established protocol. At 2, 4, 6 weeks after induction, 2g rhPDGF-B,25g Anti-PDGF-BB and saline were intramedullary injected into the bone marrow of proximal femur respectively. At week 6 after induction, distal femora were dissected for micro-CT testing. Meanwhile, the C3H10T1/2 cells were cultured with osteogenic or adipogenic differentiation medium. Meantime, the experimental cells were treated with 50 ng/ml rhPDGF-BB, 312.5 ng/ml Anti-PDGF-BB or saline. During differentiation progression, we analyzed the osteoblastic differentiation markers and mineralization, as well as the adipogenic differentiation markers and oil droplets formation and accumulation in C3H10T1/2 cells. Results: Micro-CT results showed that rhPDGF-BB could enhance the BMD, BV/TV, and Tb.Th of cancellous bone of distal femur, as well as improve the cross-sectional thickness of cortical bone in SAOP rabbit model. Meanwhile, rhPDGF-BB significantly enhanced C3H10T1/2 cells proliferation, osteogenic differentiation and calcium deposition in vitro. In contrast, the adipogenesis of C3H10T1/2 cells was suppressed in the presence of rhPDGF-BB. Conclusion: rhPDGF-BB inhibits bone loss via balancing osteogenesis and adipogenesis in SAOP rabbits.
