英文摘要利用同-PCR反应体系,分别对番茄(Lycopersicon esculentum)抗根结线虫(root-knot nematode)的Mi基因、抗番茄花叶病毒(Tomatomosaic virus)病的Tm2^2基因紧密连锁的SCAR标记进行了同时扩增筛选,扩增的特异性片段与单引物扩增片段基本完全吻合.与Mi基因紧密连锁的SCAR1标记为共显性标记,抗感材料均产生750 bp的特异片段,纯合和杂合抗病基因型存在Taq Ⅰ酶切位点,酶切后分别产生了570和200bp以及750、570和200 bp的特异性片段,而感病基因型无酶切位点;与Tm2^2基因紧密连锁的SCAR2标记也为共显性标记,抗感材料均产生950bp的特异片段,杂合抗病基因型和感病基因型有HindⅢ酶切位点,酶切后除Moperou产生800 bp特异片断外,其余杂合抗病材料均为950、500和280 bp片段.感病材料为500和280bp,纯合抗病基因型无HindⅢ酶切位点,酶切结果仍为950 bp产物.该体系可用于在同-PCR反应体系中对2个抗病基因进行同时筛选鉴定,及苗期早期辅助选育.; Mi and TM2^2 genes in tomato(Lycopersicon esculentum) tightly linked with two SCAR markers and respectively resistant to root-knot nematode and Tomato mosaic virus were amplified and screened by using a single PCR reaction. The PCR products were completely correspond to the amplified bands produced by single SCAR,primer. Among them, co-dominant SCARI marker tightly linked with Mi gene produced 750 bp fragment in both resistant and susceptible tomato lines. The amplified bands from susceptible and resistant lines were distinguishable after cleavage with the restriction enzyme Taq Ⅰ. Genotype with homozygous and heterozygous Mi gene could produce respectively 570,200 bp and 750, 570 and 200 bp bands. Susceptible genotypes still presented 750 bp fragment. The co-dominant SCAR2 marker tightly linked with Tin22 gene would produce 950 bp fragment in both resistant and susceptible tomato lines. The amplified bands from susceptible and heterozygous were distinguishable after cleavage with the restriction enzyme Hind Ⅲ. Genotype susceptible and heterozygous with Tm2^2 gene could produce respectively 500,280 bp and 950,500,280 bp bands, except line Moperous, which produced 800 bp specific fragments. Homozygous genotypes still presented 950 bp fragment. The teplicated stable results proved that two resistant genes could be identified simultaneously by using corresponding SCAR primer under adaptable condition. The established multi-PCR could be very useful for marker-assisted selection during early stage in tomato and efficiently speed up breeding procedure.
