英文摘要 | The ubiquitin proteasome system (UPS) is a highly regulated mechanism of intracellular protein degradation in eukaryotes. The UPS comprises three key enzymes:ubiquitin-activating enzyme E1, ubiquitin-conjugating enzyme E2, and ubiquitin ligase enzyme E3. There are nearly 1500 E3 ligase genes in rice and Arabidopsis genomes that target substrates for many biological processes for plant growth and tolerance to abiotic and biotic stresses. In the last decade, many studies have shown that E3 ligase genes are involved in the regulation of innate immunity in plants (Cheng and Li, 2012; Liu et al., 2012). The function of a dozen E3 ligase genes in plant immunity has recently been characterized in Arabidopsis (Furniss and Spoel, 2015; He et al., 2015). In rice, SPL11 is the first-reported U-box E3 ligase that negatively controls broad-spectrum resistance and programmed cell death in rice (Zeng et al., 2004). The RING E3 ligase OsBBI1 positively regulates resistance against Magnaporthe oryzae by modifying the rice cell wall (Li et al., 2011). Another interesting protein is RING finger E3 ligase XA21 BINDING PROTEIN 3 (XB3), which is required for the accumulation of rice bacterial blight R protein XA21 and XA21-mediated defense signaling against Xanthomonas oryzae pv. oryzae (Xoo) (Wang et al., 2006). We recently demonstrated that the fungal effector AvrPiz-t in M. oryzae suppresses PAMPtriggered immunity (PTI) by targeting the rice RING E3 ligase APIP6 for degradation (Park et al., 2012). In return, APIP6 can degrade AvrPiz-t to reduce its suppressive effects on rice PTI. Silencing of APIP6 in transgenic plants leads to reduced PTI responses and basal resistance to M. oryzae, suggesting that APIP6 positively regulates rice basal defense. Although many E3 ligase genes have been characterized in both Arabidopsis and rice, the function of their substrates is still poorly understood. In this study, we demonstrate that APIP6 interacts with and degrades OsELF3-2, an ortholog of the Arabidopsis flowering and circadian regulator ELF3. Functional analysis shows that OsELF3-2 negatively regulates immunity against M. oryzae in rice. |
