中文摘要A gene gun particle acceleration system was developed to resolve problems caused by shock waves and the microprojectile preparation in previous gene guns used for DNA immunizations. In the system, the particles coated with DNA vaccines are absorbed by a stainless steel net mounted in the acceleration channel. An experimental apparatus was designed based on the simulation results to inject pEGFG plasmids into Hela cells. The experimental results show that the particles are accelerated up to 300 m/s at an air pressure of 0.3 MPa with excellent distribution uniformity on the target plane and that the pEGFG plasmids injected into the Hela cells are well expressed, which shows that the gene gun can provide effective DNA immunization.
