The role of chemerin/CMKLR1 signaling on gestational diabetes mellitus and fetus

英文摘要Mother with obesity or gestational diabetes mellitus(GDM) often deliver large babies with increased fat mass who are susceptible to perinatal complications and to development of metabolic syndrome later in life. The serum from GDM patient or obesity pregnancy mothers was examined with higher circular chemerin, a novel adipokine dominate secreted from white fat tissue. It is currently unknown if maternal chemerin was positively related to GDM and fetal growth. First, we measured GDM patient’s serum and the result showed higher levels of chemerin compared to normal pregnancy mother. Then we compared serum level of chemerin between mice from non-pregnancy and pregnancy day 18. The result indicated chemerin levels were highly increased during pregnancy. Furthermore, the expression of chemerin and its receptors in different tissue on pregnancy day 18 demonstrated that chemerin was highly expressed in placenta and liver which indicating the secretion location of chemerin during pregnancy. At the same time, so we hypothesize that CMKLR1 signaling may play a certain role during GDM. To test our hypotheses, we constructed maternal over-nutrient and obese mice model, fed primiparous C57BL/7 mice with high fat diet during pregnancy. The chemerin in serum displayed high levels in high fat diet mice. During gestation day 12 to 16, treatment the obese mice with antagonist a-NETA, to the specific small molecule antagonist to chemerin receptor CMKLR1, improved the glucose impair demonstrated by IPGTT and fast glucose measurement. A-NETA also decreased the infant weight and infant/placenta ratio. In vitro, chemerin or CMKLR1 siRNA treatment on placenta trophoblast Bewo cell decreased protein levels for animo acid transportor protein SNAT and LAT, glucose transporter protein Glut1 and Glut3. These results indicated that chemerin may have positive correlation with GDM and large babies. CMKLR1 antagonist treatment will improve the syndrome induced by high fat diet induced obese mice, implying the pharmaceutical use of CMKLR1 small molecule compound antagonist on GDM treatment
