Tumor-Targeting and Hyaluronidase Activatable Nanoparticles for Tumor Tracer and Photochemistry Therapy

英文摘要Novel tumor-targeting hyaluronidase activatable nanoparticles have been prepared by covalent attachment of hydrophobic chlorin e6 to hyaluronic acid (HA). DOX encapsulating the nanoparticles(HA-Ce6) showed stable nano-structure(sub-100 nm size distribution)in aqueous condition and rapid uptake into tumor cells. The resulting nanoparticles HA-Ce6(DOX) could efficiently reach the tumor tissue via the passive targeting strategies and specifically enter tumor cells through the receptor-mediated endocytosis based on CD44, the HA receptor on the surface of tumor cells. Under low dose of laser power and anti-cancer drug (DOX) conditions, in vitro and in vivo studies demonstrated more effective suppression of tumor growth than free Ce6 or free DOX, which also led to overcoming the drug resistance in cancer cells. In particular, Ce6 could generate fluorescence, which showed good near-infrared fluorescence imaging in the tumor-bearing mice. Overall, it was demonstrated that HA-Ce6(DOX) nanoparticles could be successfully applied to in vivo tumor imaging and photochemistry therapy simultaneously.
