Visual tracking of Enterovirus71 infection in living mice using bioorthogonal strategy

英文摘要Enterovirus 71 (EV71) is a single-strand RNA virus, which mainly causes mild hand-foot-and-mouth disease in young children. Recently, increasing evidences have shown that EV71 infection could also be fatal due to severe systemic complications, including neurological, cardiovascular and respiratory problems [1]. However, the pathopoiesis mechanism of EV71 infection remains unclear and no effective study strategies have been developed [2]. Monitoring the in vivodynamics of EV71 is highly desirable for understanding viral pathogenesis. Herein we applied Bioorthogonal reaction to label virus with near-infrared emitting quantum dots (NIR QDs) by linking dibenzocyclooctynes (DBCO) modified viral capsids to the QDs derived with azide [3].The infection of QD-EV71 was then monitored in 3-day-old ICR mice. Mouse pups (1-3 days old) were infected with QD-EV71(1×107 pfu/mouse) by interperitoneal (i.p) injection, and the fluorescent signal of QD-EV71 was observed using an in vivo imaging system. The conjugation of QDs onto EV71 (QD-EV71) was mild and efficient without effect on viral infectivity and replication both in vitro and in vivo. After subsequent animal experiments, fluorescence signal can be observed on gut of living mice at a serial time point, paralleling the amount of virus in serum after infection. Furthermore, mice infected with QD-EV71 demonstrated bright and sustained fluorescent signals in livers, intestines, and lungs, suggesting EV71 induced pathological behaviors were governed by viral tissue tropism and distribution. However, mice administrated with free QDs only showed weak signals in livers. Especially in lung tissues, the fluorescent signals of QD-EV71 group were 2 folds higher than that of QD group. The different distribution of QD-EV71 and free QDs should be due to the systemic infection caused by QD-EV71.
