英文摘要 | Gene therapy can be defined as the transfer of genetic material into a cell for therapeutic purposes. Cytosine deaminase (CD) transferred into tumor cells by an adenoviral vector, can convert the antifungal drug fluorocytosine (5-FC) to the antimetabolite 5-fluorouracil (5-FU), which kills not only the transfected tumor cells but also their neighbors by the so-called ‘bystander effect’. Here, we prepared polypeptide micelle nanoparticles (NP) of a triblock copolymer poly(ethylene glycol)-b-poly(l-lysine)-b-poly(l-leucine) (PEG-PLL-PLLeu) to systemically deliver CD plasmid for an effective gene vector. The promoter sequences of the gene that are highly active in hepatoma tumor cells, HE4, were used to target HepG2 tumor cells. We constructed the CD plasmid and successfully transfected into HepG2 tumor cells. The transfection efficiency of CD suicide gene complexes in HepeG2 cells was 73.1%. This suicid gene delivery system can significantly inhibited HepeG2 tumor cell proliferation. Our results demonstrated well-defined PEG-PLL-PLLeu polypeptide cationic micelles with the excellent gene delivery of CD plasmid and 5-FC in combined cancer therapy, which holds promise as an effective therapy for advanced-stage hepatoma cancer. |
